The p38 MAPK cascade, also known as the p38 mitogen-activated protein kinase pathway, is a crucial signaling pathway involved in cellular responses to stress and inflammation. It is part of the larger family of MAP kinases, which are involved in regulating a wide variety of cellular processes including cell growth, differentiation, and survival.

The p38 MAPK cascade is activated by a range of extracellular stimuli such as cytokines (like TNF-alpha and IL-1), growth factors, ultraviolet radiation, and osmotic shock. Consequently, the p38 MAPK pathway plays a significant role in mediating inflammatory responses, immune cell activation, and cellular stress responses. Dysregulation of the p38 MAPK cascade has been implicated in various diseases, including inflammatory disorders like rheumatoid arthritis, cardiovascular diseases, and cancer. Therefore, it represents a potential target for therapeutic interventions.
