A luminometer is a scientific instrument used to measure luminescence. Luminescence is the emission of light by a substance not resulting from heat. This includes phenomena such as chemiluminescence, bioluminescence, fluorescence, and phosphorescence. Luminometers are designed to detect and quantify the light produced by these processes.

The basic principle of operation involves a photosensitive detector, such as a photodiode or photomultiplier tube, positioned to receive light emitted from a sample. The sample, often a liquid in a cuvette or a solid in a petri dish, is placed in close proximity to the detector. An excitation source, if required for fluorescence or phosphorescence measurements, is also incorporated into the instrument. The intensity of the emitted light is then converted into an electrical signal, which is amplified and displayed as a numerical value, typically in units of relative light units (RLU).

Luminometers find widespread use in various scientific disciplines. In biology and medicine, they are crucial for assays involving ATP detection, which indicates the presence of viable cells or microbial contamination. They are also employed in reporter gene assays, DNA probe assays, and enzyme activity measurements. In environmental science, luminometers can be used to assess water quality by measuring bioluminescent bacteria. The sensitivity and specificity of luminometers make them valuable tools for detecting low levels of light-emitting substances.
