Heparinase is an enzyme that catalyzes the degradation of heparin and heparan sulfate. These are complex carbohydrates belonging to the glycosaminoglycan family, known for their anticoagulant properties and involvement in various cellular processes. Heparinase enzymes are classified as heparin lyases.

There are several types of heparinases, primarily heparinase I, heparinase II, and heparinase III, distinguished by their substrate specificities and cleavage mechanisms. Heparinase I cleaves heparin and heparan sulfate at internal glucosamine-sulfate bonds. Heparinase II can cleave both internal and terminal linkages, and heparinase III primarily acts on N-sulfoglucosamine bonds. These enzymes are typically found in specific bacteria, such as Flavobacterium heparinum, and are produced as part of their metabolic machinery to break down extracellular matrix components.

The primary application of heparinase is in research settings. It is used to depolymerize heparin and heparan sulfate chains, allowing for the analysis of their structures and functions. This enzymatic degradation is crucial for understanding the roles of these glycosaminoglycans in biological processes like cell growth, differentiation, viral entry, and inflammation. Therapeutically, heparinase has been explored as an antagonist to heparin's anticoagulant effects, potentially for reversing excessive anticoagulation caused by heparin overdose or during specific surgical procedures. However, its clinical use is limited due to factors like immunogenicity and delivery challenges.
