A folate analog is a chemical compound that structurally resembles folic acid, also known as vitamin B9. Folic acid is essential for numerous bodily functions, including DNA synthesis and repair, as well as cell division and growth. Folate analogs can interfere with these processes by competing with folic acid for binding to enzymes involved in folate metabolism.

In biological systems, folic acid is converted into its active form, tetrahydrofolate (THF), which serves as a cofactor for enzymes that transfer one-carbon units. These one-carbon transfers are critical for the synthesis of purines and pyrimidines, the building blocks of DNA and RNA. They are also important for the metabolism of amino acids, such as serine and methionine.

Folate analogs can exert their effects by inhibiting the enzymes that process folic acid, thereby depleting the cell's supply of active folate cofactors. This inhibition can disrupt DNA synthesis, leading to cell death, particularly in rapidly dividing cells. Because cancer cells often divide more rapidly than normal cells, folate analogs have found significant application in cancer chemotherapy. Methotrexate is a well-known example of a folate analog used to treat various cancers, including leukemia, lymphoma, and breast cancer. It works by inhibiting dihydrofolate reductase, a key enzyme in folate metabolism. Other folate analogs, such as pemetrexed, are also used in cancer treatment.

Beyond cancer therapy, folate analogs can be used to study folate metabolism and its role in various biological processes. They can also be used to induce folate deficiency in experimental models. However, their interference with essential metabolic pathways means they can also have toxic side effects, especially on rapidly dividing normal tissues like bone marrow and the gastrointestinal tract.
