fasberoende

Fasberoende is a Swedish term used in biomedical contexts to describe a dependence on Fas signaling, particularly in relation to the extrinsic apoptotic pathway. The concept refers to cellular or tissue processes in which apoptosis or cell fate is primarily driven by Fas (CD95) receptor engagement and the downstream signaling cascade. The term appears mainly in specialized literature and is not widely used outside of immunology and cell biology discussions.

Mechanism and pathway. When Fas ligand (FasL) binds to the Fas receptor, the death-inducing signaling complex (DISC) forms and recruits adaptor proteins such as FADD, leading to activation of initiator caspase-8. This initiates a caspase cascade that drives apoptosis. In some cells, caspase-8 can cleave Bid to amplify the death signal through the mitochondria, linking the extrinsic pathway with the intrinsic pathway. Modulators such as cFLIP, decoy receptors, and various inhibitors can influence Fas-dependent signaling, tipping the balance toward survival or death.

Biological role. Fas signaling plays a key role in immune system regulation, including deletion of autoreactive cells and maintenance of peripheral tolerance. It also contributes to tissue turnover and development in certain contexts. Dysregulation of Fasberoende processes can contribute to autoimmune disease, immune evasion by cancer cells, or excessive tissue damage if Fas signaling is aberrantly activated.

Clinical and research relevance. Because Fas-mediated apoptosis is a potential route to eliminate diseased cells, researchers study Fas agonists and antagonists as therapeutic strategies. Challenges include context-dependent responses and potential toxicity, for example to the liver, which has shown sensitivity to Fas-targeted approaches.
