Dentinopredentin is a thin, unmineralized layer of predentin that lies adjacent to the dental pulp. It is the most recently formed organic matrix of dentin and is located immediately beneath the odontoblast layer, which are the cells responsible for dentin formation. Predentin is essentially newly secreted dentin matrix before it undergoes mineralization.

The process of dentinogenesis involves the differentiation of odontoblasts from the dental papilla. These odontoblasts then begin to synthesize and secrete an organic matrix composed primarily of collagen type I. This matrix is deposited at the pulpal boundary and gradually mineralizes to form mature dentin. The unmineralized layer at the very edge of the pulp is what is termed predentin.

Predentin plays a crucial role in the continuous formation and apposition of dentin throughout life. As odontoblasts secrete more matrix, the predentin layer thickens, and the newly formed matrix mineralizes, pushing the odontoblast layer further away from the pulp. This ongoing process contributes to the overall growth and repair of the tooth. The presence of predentin is a characteristic feature of developing and continuously forming dentin. Its unmineralized state allows for the diffusion of nutrients and signaling molecules from the pulp to the mineralizing dentin.
