Calcitonin receptor, also known as CALCR, is a cell surface receptor that binds to calcitonin and calcitonin gene-related peptide (CGRP). It is a seven-transmembrane G protein-coupled receptor. The gene encoding the calcitonin receptor is located on chromosome 11 in humans. Calcitonin receptor plays a crucial role in calcium and bone metabolism. Upon binding of calcitonin, it activates adenylyl cyclase, leading to an increase in intracellular cyclic AMP levels. This signaling pathway ultimately results in the inhibition of osteoclast activity, which are cells responsible for bone resorption. Consequently, calcitonin receptor activation contributes to a decrease in blood calcium levels and a reduction in bone breakdown.

Calcitonin receptor is expressed in various tissues, including bone, kidney, and the brain. In bone, its presence on osteoclasts is vital for the hypocalcemic and bone-protective effects of calcitonin. In the kidney, it influences calcium reabsorption. In the central nervous system, its role is less understood but may be involved in pain perception and other neurological functions. Dysregulation of calcitonin receptor signaling has been implicated in conditions such as osteoporosis and Paget's disease of bone.
