Acylcoenzyme refers to a group of molecules that play a crucial role in cellular metabolism, particularly in energy production and biosynthesis. These molecules are characterized by the presence of an acyl group attached to Coenzyme A (CoA). Coenzyme A is a vital coenzyme derived from pantothenic acid (vitamin B5) and is involved in a multitude of enzymatic reactions.

The acyl group is a functional group derived from a carboxylic acid by removal of its hydroxyl group. In acylcoenzymes, this acyl group is covalently linked to the sulfhydryl group of Coenzyme A. This thioester linkage is high-energy, meaning that the hydrolysis of this bond releases a significant amount of free energy. This energy is often harnessed by cells to drive other biochemical reactions.

The most well-known and abundant acylcoenzyme is acetyl-CoA. Acetyl-CoA is a central molecule in metabolism, formed from the breakdown of carbohydrates, fats, and proteins. It enters the citric acid cycle (also known as the Krebs cycle), a key pathway for ATP generation. Other important acylcoenzymes include succinyl-CoA, propionyl-CoA, and malonyl-CoA, each involved in specific metabolic pathways such as the synthesis of heme, fatty acids, and cholesterol. The interconversion and utilization of these acylcoenzymes are tightly regulated to maintain cellular homeostasis.
