Vitellinase

Vitellinase is an enzyme that plays a critical role in the breakdown of vitellin, a major yolk protein found in the eggs of vertebrates, particularly birds and reptiles. Vitellin is a precursor to lipovitellin, a phosphoglycoprotein that stores lipids and phosphorus within the yolk, providing essential nutrients for embryonic development. Vitellinase facilitates the hydrolysis of vitellin into smaller peptides and amino acids, which are then utilized by the developing embryo.

This enzyme is primarily produced by the embryo itself during early stages of development, allowing it to access the nutrients stored in the yolk. The process of vitellin degradation is tightly regulated to ensure that the embryo receives a steady supply of nutrients while minimizing waste. Vitellinase activity is particularly important in species where the embryo relies entirely on yolk reserves for sustenance before hatching or birth.

Some tumor cells, including those in ovarian and breast cancers, express vitellinase-like activity, suggesting a potential role in nutrient acquisition for rapidly dividing cancer cells. This has led to investigations into whether inhibitors of vitellinase could be developed as therapeutic agents to disrupt tumor growth by limiting their access to essential nutrients.

The enzyme is classified as a metalloprotease, meaning it requires a metal ion, typically zinc, for its catalytic activity. Its structure and function have been characterized in various species, including chickens, quails, and fish, where it serves a similar role in yolk utilization during embryonic development.
