TrxRs

Thioredoxin reductases (TrxRs) are a family of enzymes that play a crucial role in cellular redox homeostasis. They are part of the larger thioredoxin system, which also includes thioredoxin (Trx) and thioredoxin peroxidases (Tpx). TrxRs catalyze the reduction of oxidized thioredoxin, thereby regenerating its active reduced form. This reduced thioredoxin is a potent biological antioxidant and a key component in numerous cellular processes, including DNA synthesis and repair, protein folding, and immune responses.

The thioredoxin reductase enzyme typically utilizes NADPH as a reducing cofactor. The catalytic mechanism involves a CXXC active site motif within the thioredoxin reductase, which is essential for the transfer of electrons from NADPH to the thioredoxin substrate. Different organisms possess various isoforms of TrxRs, with distinct tissue specificities and cellular localizations, reflecting their diverse roles. For example, mammalian cells have at least three TrxR isoforms with varying functions.

TrxRs are involved in the redox regulation of a wide range of proteins and play a significant role in cellular signaling pathways. Dysregulation of TrxR activity has been implicated in various diseases, including cancer, neurodegenerative disorders, and inflammatory conditions. Consequently, TrxRs have emerged as potential therapeutic targets, with research focusing on developing inhibitors or modulators of their activity for the treatment of these diseases.
