Thioxanthenes

Thioxanthenes are a class of organic compounds characterized by a tricyclic structure consisting of a benzene ring fused to a thioxanthene ring, which is itself a sulfur analog of xanthene. The core thioxanthene structure is dibenzothiopyran. This structural motif is significant in medicinal chemistry, particularly as the basis for a group of antipsychotic drugs known as thioxanthene antipsychotics.

These thioxanthene antipsychotics, such as chlorprothixene, flupentixol, and zuclopenthixol, act primarily by blocking dopamine receptors in the brain, specifically dopamine D1 and D2 receptors. Their pharmacological effects are similar to those of phenothiazine antipsychotics, though with some differences in their side effect profiles. They are used in the management of psychotic disorders like schizophrenia and bipolar disorder. The specific substituents on the thioxanthene ring and the side chain attached to the sulfur atom influence their potency, receptor selectivity, and pharmacokinetic properties.
