RanGTPasesystemet, also known as the Ran GTPase cycle, is a critical cellular mechanism involved in the regulation of nucleocytoplasmic transport, mitosis, and nuclear envelope assembly. It centers around the Ran protein, a small GTP-binding protein, which functions as a molecular switch cycling between its GTP-bound active form and GDP-bound inactive form.

The RanGTPase system maintains directional transport of proteins and RNA between the nucleus and cytoplasm. In the nucleus, Ran is predominantly in its GTP-bound state, facilitated by the guanine nucleotide exchange factor RCC1. In the cytoplasm, Ran is predominantly hydrolyzed to its GDP-bound form, aided by RanGAP. This gradient of Ran-GTP and Ran-GDP across the nuclear envelope provides the necessary directional cues for nuclear import and export receptors, such as importins and exportins.

During nuclear import, cargo complexes bind importins in the cytoplasm and translocate through the nuclear pore complex (NPC). Once inside the nucleus, Ran-GTP binds to importins, inducing the release of their cargo. Conversely, for nuclear export, Ran-GTP associates with exportins and their cargo within the nucleus; upon entering the cytoplasm, Ran hydrolyzes GTP to GDP, prompting cargo release.

Beyond transport, the RanGTPase system plays a vital role in spindle assembly during mitosis and re-establishment of the nuclear envelope after cell division. Its regulation ensures proper cell cycle progression and genomic stability.

Overall, the RanGTPase system is indispensable for maintaining cellular compartmentalization and ensuring accurate gene expression and cell division, highlighting its significance in cellular physiology.
