Northern blot analysis is a molecular biology technique used to detect specific RNA molecules within a sample. It is an essential tool for studying gene expression, as it allows researchers to determine the presence, quantity, and size of RNA transcripts.

The process begins with the isolation of RNA from cells or tissues. This RNA is then separated based on size by gel electrophoresis, typically using an agarose gel. The separated RNA molecules are subsequently transferred from the gel onto a solid membrane, such as a nitrocellulose or nylon membrane. This transfer is the "blotting" step.

Following the transfer, the membrane is incubated with a labeled probe. This probe is a short sequence of nucleotides that is complementary to the target RNA molecule. The probe can be labeled with radioactive isotopes or non-radioactive tags, such as fluorescent molecules. The probe hybridizes (binds) specifically to its complementary RNA sequence on the membrane.

Finally, the presence of the hybridized probe is detected using appropriate methods, depending on the label used. This detection reveals the location and amount of the target RNA. By comparing the signal intensity to known standards, researchers can quantify the abundance of the RNA. Northern blotting is valuable for analyzing alternative splicing, RNA degradation, and the developmental or tissue-specific expression of genes.
