MOB1dependent refers to a class of biological processes and signaling pathways that rely on the activity of the MOB1 (Mps One Binder) protein family. MOB1 proteins are highly conserved across eukaryotes, playing essential roles in cell cycle regulation, mitosis, and other critical cellular functions.

The MOB1 family includes two main members, MOB1A and MOB1B, which function as regulatory subunits for various kinases. Their primary role is to activate the Large Tumor Suppressor kinase 1 (LATS1) and LATS2, which are central components of the Hippo signaling pathway. This pathway is crucial for controlling organ size, cell proliferation, and apoptosis. By binding to and stabilizing LATS kinases, MOB1 proteins facilitate their phosphorylation of downstream targets such as Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ). This phosphorylation leads to their cytoplasmic retention or degradation, thereby suppressing cell proliferation and promoting tissue homeostasis.

Beyond the Hippo pathway, MOB1 proteins also interact with other kinases, including the NDR (nuclear Dbf2-related) kinases, which are involved in cell cycle progression and cytoskeletal organization. MOB1’s role in these processes underscores its importance in maintaining cellular integrity and preventing uncontrolled growth, which is often dysregulated in cancer.

Mutations or dysregulation of MOB1 have been implicated in various diseases, including cancer, where its loss can lead to aberrant cell proliferation.
