Luciferinsubstrat

Luciferinsubstrat is a term encountered in some multilingual or informal sources to denote the substrate molecules that participate in bioluminescent reactions catalyzed by luciferase enzymes. In standard English usage, the substrate is typically called luciferin, and luciferin itself is consumed during light-emitting oxidation. Consequently, luciferinsubstrat is not widely used in primary literature, but may appear as a direct translation or variant of "luciferin substrate."

Bioluminescent reactions vary by system. In the common firefly (Photinus and Luciola) luciferase system, D-luciferin reacts with ATP and molecular oxygen in the active site, forming luciferyl-adenylate, which oxidizes to oxyluciferin with concomitant emission of photons. The emitted light typically peaks in the green to yellow range (approximately 550–570 nm) and depends on enzyme source, substrate analogs, and reaction conditions.

Other luciferin substrates include coelenterazine, used by many marine luciferases such as Renilla and Gaussia, and bacterial luciferin, which requires FMNH2, an aldehyde substrate, and oxygen. Researchers also synthesize luciferin analogs or engineered luciferases to modify brightness, emission color, or substrate affinity to suit imaging or assay needs.

Applications of bioluminescent substrates span live-cell and in vivo imaging, gene expression reporting, promoter activity studies, and high-throughput screening. The choice of substrate affects sensitivity, emission color, and tissue penetration, with D-luciferin being the standard for firefly luciferase imaging in mammals.
