Lineweaver-Burk plot, named for Hans Lineweaver and Dean Burk, is a graphical method used in enzyme kinetics to linearize the Michaelis-Menten relationship and simplify the estimation of kinetic constants. It remains a classic teaching tool and a historical reference in enzymology.

The plot is formed by graphing the reciprocal of the reaction velocity (1/v) on the y-axis against the reciprocal of the substrate concentration (1/[S]) on the x-axis. The linear form of the Michaelis-Menten equation is 1/v = (Km/Vmax)(1/[S]) + 1/Vmax, where the slope equals Km/Vmax, the y-intercept equals 1/Vmax, and the x-intercept equals -1/Km. This allows Km and Vmax to be read from the best-fit line.

Lineweaver-Burk plots are useful for comparing enzyme kinetics under different conditions or in the presence of inhibitors, where multiple lines can illustrate changes in Km and/or Vmax. However, the method has drawbacks. It disproportionately weights data at low substrate concentrations, amplifying experimental error and potentially biasing estimates. Because of these issues, nonlinear regression directly fitting the Michaelis-Menten equation to the original data is often preferred in modern analyses.

In practice, the Lineweaver-Burk plot remains a valuable educational tool and a historical reference in kinetic studies, offering a straightforward, if imperfect, means to visualize relationships among v, [S], Km, and Vmax and to gain initial intuition about enzyme behavior and inhibition patterns.
