Kinasiinipohjaiset

Kinasiinipohjaiset is a Finnish term used to describe substances, therapies, or approaches that are based on or target kinases. In biomedical literature, the concept covers strategies that influence kinase enzymes, which regulate phosphorylation signaling pathways that control cell growth, differentiation, and survival.

Kinase-based therapies are primarily built on kinase inhibitors, usually small molecules that bind to the catalytic domain of kinases and block phosphorylation. Some inhibitors are ATP-competitive, while others act allosterically or through other mechanisms. Target kinases include receptor tyrosine kinases (for example EGFR, VEGFR, ALK), non-receptor tyrosine kinases (such as BCR-ABL, SRC), and serine/threonine kinases (including PI3K, AKT, mTOR, CDKs). Well-known examples of kinase inhibitors include imatinib, gefitinib, erlotinib, sunitinib, sorafenib, and lapatinib.

Clinical use and challenges: Kinase inhibitors are widely used in oncology, hematology, and inflammatory diseases due to their ability to selectively disrupt aberrant signaling. Benefits can include targeted action and improved tolerability compared with traditional chemotherapy. Challenges include acquired resistance driven by mutations or pathway redundancy, as well as adverse effects like rash, hypertension, cytopenias, and organ-specific toxicities.

Research directions: Ongoing development focuses on next-generation inhibitors with greater selectivity and resistance-evading properties, covalent and allosteric inhibitors, and approaches to degrade kinases using technologies such as PROTACs. Combination therapies and precision medicine aim to tailor kinase-based interventions to individual patient profiles.
