Glycosphingolipids are a class of complex lipids that consist of a ceramide backbone with one or more sugar residues attached. They are integral components of animal cell membranes, especially on the outer leaflet of the plasma membrane, where they contribute to membrane structure and organization.

The main structural categories are neutral glycosphingolipids and acidic glycosphingolipids. Neutral glycosphingolipids include cerebrosides (glucosylceramide or galactosylceramide) and more elaborate globosides that carry several sugar units. Acidic glycosphingolipids are primarily gangliosides, which contain sialic acid and can present complex oligosaccharide chains. The diversity of glycosphingolipids arises from different sugar compositions and linkages, producing numerous individual species such as GM1, GM2, GD1a, and Gb3.

Biosynthesis occurs in the Golgi apparatus, where glycosyltransferase enzymes add sugars to ceramide in a stepwise fashion. The resulting glycosphingolipids are delivered to the cell surface via vesicular trafficking. Their distribution in membrane microdomains, including lipid rafts, allows them to participate in cell–cell recognition, adhesion, and signaling. Gangliosides, in particular, play prominent roles in the nervous system and modulate receptor activity and synaptic function.

Clinical relevance involves metabolic disorders and disease associations. Defects in lysosomal degradation of glycosphingolipids lead to storage diseases, such as Fabry disease (accumulation of globotriaosylceramide) and GM2 gangliosidoses. Altered glycosphingolipid metabolism and distribution have also been linked to neurodegenerative conditions and cancer biology.
