Esteröitymismekanismeihin, the processes by which esters are formed from carboxylic acids and alcohols, are fundamental to organic chemistry. The most common pathway is the Fischer esterification, a reversible acid‑catalysed reaction in which a carboxylic acid protonates the carbonyl oxygen, increasing electrophilicity. An alcohol nucleophile then attacks the carbonyl carbon, forming a tetrahedral intermediate. Proton transfer and elimination of water subsequently regenerate the carbonyl and yield the ester. The reaction equilibrium is shifted toward product formation by removing water or using excess alcohol.

Catalysts other than strong acids can also drive esterification. Lewis acids such as zinc chloride or boron trifluoride coordinate to the carbonyl oxygen, lowering the activation barrier and enabling lower temperature conditions. Enzymatic esterification, performed by lipases, proceeds through a catalytic triad mechanism that mimics the acid‑catalysed pathway but operates under mild, aqueous conditions.

Esterification can also occur via alcoholysis of acid chlorides, acyl anhydrides, or mixed anhydrides, which use more reactive acylating agents. These methods bypass the equilibrium limitation of the Fischer route, allowing high yields in a single step. In all mechanisms, the key mechanistic features are nucleophilic attack on an activated carbonyl and subsequent elimination of a leaving group, which collectively convert a carboxylate group into an ester linkage.
