Endothelinisoformit refers to the different forms or isoforms of endothelin, a family of potent vasoconstricting peptides. The primary endothelin genes encode for three distinct isoforms: Endothelin-1 (ET-1), Endothelin-2 (ET-2), and Endothelin-3 (ET-3). These isoforms are produced by a variety of cells and play crucial roles in physiological processes.

Endothelin-1 is the most well-characterized isoform and is primarily produced by endothelial cells. It is a key regulator of vascular tone, influencing blood pressure and blood flow. ET-1 also has roles in cell growth, differentiation, and tissue repair. Endothelin-2 is mainly found in the kidneys and intestines, and its functions are less understood but are thought to be related to epithelial cell function. Endothelin-3 is widely distributed throughout the body and is particularly important during embryonic development, playing a role in neural crest cell migration and the development of the enteric nervous system.

The production of endothelin isoforms is tightly regulated. They are synthesized as larger precursor molecules called preproendothelin, which are then processed into biologically active peptides. These peptides exert their effects by binding to specific endothelin receptors, ET_A and ET_B, which are present on various cell types. The differential expression and signaling of these receptors contribute to the diverse physiological actions of endothelin isoforms. Dysregulation of endothelin signaling has been implicated in various diseases, including hypertension, pulmonary hypertension, and cardiovascular disease.
