DPPHtestillä is a Finnish term that translates to "with the DPPH test" or "using the DPPH test". The DPPH test itself is a widely used laboratory method for evaluating the antioxidant capacity of a substance. DPPH stands for 2,2-diphenyl-1-picrylhydrazyl, which is a stable free radical that exhibits a deep purple color. When an antioxidant compound is present, it donates an electron to the DPPH radical, neutralizing it. This neutralization causes a color change from purple to yellow. The extent of this color change, measured spectrophotometrically, is directly proportional to the antioxidant activity of the substance being tested.

In practice, the DPPH test involves dissolving DPPH in a suitable solvent, typically an organic solvent like methanol or ethanol. The sample containing potential antioxidants is then added to this solution. After a set incubation period, the absorbance of the mixture is measured at a specific wavelength, usually around 517 nm. A decrease in absorbance indicates that the DPPH radical has been scavenged by the antioxidants in the sample. The results are often expressed as an IC50 value, which represents the concentration of the sample required to scavenge 50% of the DPPH radicals. This test is frequently employed in food science, natural product research, and pharmaceutical studies to compare the antioxidant potential of various extracts, compounds, and food products.
