Acetogenins

Acetogenins are a class of naturally occurring secondary metabolites primarily isolated from plants in the order *Laurales*, particularly from the families *Lauraceae* and *Annonaceae*. These compounds are characterized by their polyketide origin and often exhibit complex structures containing multiple acetogenic units, typically featuring long carbon chains with various functional groups such as hydroxyls, ketones, and epoxides. Their biosynthesis involves the condensation of acetate units, which gives rise to their name.

Acetogenins are renowned for their diverse biological activities, including potent cytotoxic, antimicrobial, and anti-parasitic properties. Some members of this class have shown promise in cancer research due to their ability to inhibit mitochondrial function by targeting the electron transport chain, particularly complex I. This mechanism disrupts cellular respiration, leading to apoptosis in cancer cells. Notable examples include annonacin and bullatacin, which have been studied extensively for their cytotoxic effects.

Chemically, acetogenins are often classified based on their structural features, such as the presence of a tetrahydrofuran ring or a macrolide lactone. Their synthesis in plants is believed to occur through a polyketide pathway, followed by enzymatic modifications that introduce functional diversity. Due to their complexity, acetogenins have posed significant challenges for total synthesis in the laboratory, though advancements in synthetic chemistry have enabled the production of some analogs.

Despite their potential therapeutic applications, acetogenins also exhibit toxicity, particularly neurotoxicity, which has limited their clinical use.
