Tropokollageenit are the precursor molecules that assemble to form collagen fibers. They are rod-shaped proteins approximately 300 nanometers long and 1.5 nanometers in diameter. Each tropokollagen molecule consists of three polypeptide chains, known as alpha chains, which are wound around each other in a helical structure. These chains are rich in the amino acids glycine, proline, and hydroxyproline.

The synthesis of tropokollagen begins within cells, primarily fibroblasts. After synthesis, these molecules undergo post-translational modifications, including hydroxylation of proline and lysine residues, and glycosylation. These modifications are crucial for the stability and function of the final collagen structure. Once synthesized and modified, tropokollagen molecules are secreted from the cell into the extracellular space.

In the extracellular environment, tropokollagen molecules spontaneously self-assemble into larger fibrils. This assembly process involves a staggered arrangement of tropokollagen units, which creates a characteristic banding pattern observed in mature collagen fibers under electron microscopy. Cross-linking between tropokollagen molecules then occurs, further strengthening the collagen fibrils and providing mechanical integrity to connective tissues. Tropokollagen assembly and cross-linking are essential steps in the formation of robust and functional collagen, which provides structural support to skin, bones, tendons, and other tissues. Defects in tropokollagen formation or assembly can lead to various connective tissue disorders.
