Superantigens are a class of potent immune stimulants that trigger a disproportionately strong response from the immune system. Unlike conventional antigens that require specific recognition by T cells and antigen-presenting cells, superantigens can bind to both the major histocompatibility complex (MHC) class II molecule on antigen-presenting cells and the T cell receptor (TCR) on T cells simultaneously. This cross-linking bypasses the normal antigen-specific activation pathway.

This non-specific binding leads to the activation of a vast number of T cells, often a significant percentage of the entire T cell population. This massive T cell proliferation results in the release of a large quantity of cytokines, a phenomenon known as a cytokine storm. The excessive cytokine production can cause widespread inflammation, tissue damage, and can be life-threatening, leading to symptoms like fever, rash, hypotension, and organ failure.

Superantigens are often produced by microorganisms, including bacteria and viruses. For example, certain strains of *Staphylococcus aureus* and *Streptococcus pyogenes* produce superantigens responsible for conditions like toxic shock syndrome and scarlet fever. Understanding superantigens is crucial for developing strategies to mitigate their harmful effects and to better comprehend the complex interactions between pathogens and the immune system.
