Proteoglycans are synthesized in the rough endoplasmic reticulum and then modified in the Golgi apparatus before being secreted into the extracellular space. Their structure and function can be regulated by various post-translational modifications, including sulfation, acetylation, and O-glycosylation. Abnormalities in proteoglycan structure or function have been linked to several diseases, including osteoarthritis, Ehlers-Danlos syndromes, and certain forms of cancer.
