This reaction is a crucial step in the catalytic cycle of several multi-enzyme complexes involved in intermediary metabolism. The primary enzymes responsible for this reduction are dihydrolipoamide dehydrogenases (also known as Lpd), which are flavoproteins that utilize NAD+ as a cofactor.

The process involves the transfer of a hydride ion from NADH to the disulfide bond of lipoamide, breaking the S-S bond and forming two sulfhydryl groups in dihydrolipoamide.
