Acrystallins are a family of proteins that, in vertebrates, are primarily found in the lens of the eye. Their name derives from their function as structural components of the lens, contributing to its transparency and refractive properties. While initially thought to be exclusively lens-specific, acrystallins have since been identified in other tissues and even in invertebrates, suggesting broader roles.

The most well-characterized acrystallins are alpha-, beta-, and gamma-crystallins. These proteins are highly soluble and tend to aggregate into specific oligomeric structures within the lens. This ordered arrangement is crucial for minimizing light scattering, allowing for clear vision. Mutations in genes encoding crystallins have been linked to various forms of cataracts, a condition characterized by clouding of the lens.

Beyond their structural role, research has uncovered potential functional activities for acrystallins. Some have exhibited chaperone-like properties, assisting in protein folding and preventing aggregation of other cellular proteins. Others have been implicated in antioxidant defense and UV protection. The discovery of acrystallin-like proteins in non-vertebrate organisms suggests that their evolutionary origins and functional diversity may be more extensive than previously understood. Their presence in diverse species indicates fundamental biological importance beyond the vertebrate eye lens.
