**rRNA Depletion**

Ribosomal RNA (rRNA) depletion is a technique used in molecular biology to selectively remove ribosomal RNA from total RNA samples, allowing for the isolation of other smaller RNA species such as microRNAs (miRNAs), small nuclear RNAs (snRNAs), and small cytoplasmic RNAs. This process is particularly valuable in studies requiring detailed analysis of non-coding RNAs, as rRNA constitutes the majority of cellular RNA, overwhelming smaller transcripts in sequencing libraries.

The method typically involves binding rRNA to immobilized oligonucleotides complementary to conserved regions of 18S and 28S rRNA (in eukaryotes) or 16S and 23S rRNA (in prokaryotes) using hybridization and magnetic beads or streptavidin-coated particles. After washing away unbound RNA, the rRNA-bound beads are separated, and the remaining RNA—now enriched for smaller transcripts—is eluted for downstream applications such as RNA sequencing (RNA-seq), microarray analysis, or Northern blotting.

Advantages of rRNA depletion include improved sequencing depth for low-abundance RNAs, reduced background noise in high-throughput assays, and enhanced detection of regulatory RNAs like miRNAs, which are often masked by rRNA in untreated samples. However, the technique requires careful optimization to minimize loss of other RNA classes, particularly if high-fidelity recovery is needed. Alternative approaches, such as poly(A) tail enrichment or size selection, may be used in conjunction with rRNA depletion to further refine RNA fractions.

Common reagents and kits for rRNA depletion include those from companies like Illumina, Bio-Rad, and Ambion, which provide standardized protocols for eukaryotic and prokaryotic systems. Proper handling of RNA samples, including RNA integrity checks and precise depletion conditions, is essential to ensure reproducibility and accuracy in experimental results.
