RPE65dependent refers to a biological process or pathway that relies on the protein RPE65 for its function. RPE65 is a key enzyme found in the retinal pigment epithelium (RPE), a layer of cells in the back of the eye that supports the photoreceptor cells responsible for vision.

The primary role of RPE65 is in the visual cycle, a biochemical process essential for regenerating the light-sensitive molecule called rhodopsin in the photoreceptors. When light strikes rhodopsin, it triggers a cascade of events that ultimately leads to a visual signal being sent to the brain. After this signal is initiated, rhodopsin is chemically altered and needs to be restored to its original form to be able to detect light again. This regeneration process involves a series of enzymatic reactions, and RPE65 plays a crucial role in one of the rate-limiting steps.

Therefore, RPE65dependent processes are those that cannot occur or are severely impaired without the activity of this specific protein. Mutations or deficiencies in the RPE65 gene can lead to a group of inherited retinal diseases, collectively known as Leber congenital amaurosis (LCA) and retinitis pigmentosa (RP). These conditions are characterized by severe vision loss from birth or early childhood, as the inability to regenerate rhodopsin disrupts the continuous functioning of the visual system. Understanding RPE65dependent pathways is therefore vital for research into the diagnosis and potential therapeutic interventions for these blinding inherited eye disorders. Gene therapy approaches targeting the RPE65 gene are currently being investigated and developed as treatments for some of these conditions.
