PI3KPIP3

PI3KPIP3, also known as phosphatidylinositol-3,4,5-trisphosphate, is a lipid molecule that plays a crucial role in cellular signaling. It is a phosphorylated derivative of phosphatidylinositol 4,5-bisphosphate (PIP2) and is found in the inner leaflet of the plasma membrane. PI3KPIP3 is synthesized by the enzyme phosphoinositide 3-kinase (PI3K). Its production is a key event in the activation of the PI3K/Akt signaling pathway, which is involved in a wide range of cellular processes.

The primary function of PI3KPIP3 is to serve as a docking site for proteins that contain pleckstrin homology (PH) domains. Many of these PH-domain-containing proteins are critical components of signaling cascades, including kinases like Akt, phosphoinositide-dependent kinase-1 (PDK1), and guanine nucleotide exchange factors for small GTPases. By recruiting these proteins to the membrane, PI3KPIP3 facilitates their activation and subsequent downstream signaling. This pathway is essential for cell growth, survival, proliferation, and metabolism. Dysregulation of PI3KPIP3 levels and the PI3K/Akt pathway are implicated in various diseases, most notably cancer, where uncontrolled cell growth is a hallmark.
