Mikroskoopit

Mikroskoopit are instruments that magnify small objects beyond the naked eye, enabling observation of cellular structures, microorganisms, and material features. The term mikroskoopit is Finnish for microscopes. They range from simple optical devices to advanced imaging systems used in science, medicine, and engineering.

Microscopy began with simple lenses in the 16th–17th centuries. Antonie van Leeuwenhoek and Robert Hooke observed bacteria, cells, and fungi, advancing biological imaging. The 19th century saw improvements to the compound light microscope, increasing resolution. The 20th century introduced the electron microscope (first practical model by Ernst Ruska in 1931), enabling ultrastructural imaging.

Modern microscopes rely on optics or electron beams. Key concepts include magnification, resolution, and numerical aperture. Common components are the objective lens, eyepiece, stage, condenser, and illumination system; sample preparation and contrast techniques such as phase contrast or DIC improve visibility.

Light microscopes use visible light and techniques such as fluorescence, phase contrast, or confocal optics to study living cells and materials at modest resolutions. Electron microscopes achieve higher resolution: scanning electron microscopes reveal surface topography, while transmission electron microscopes show internal structure. Scanning probe microscopes, including atomic force microscopy, measure surface properties at the nanoscale.

Limitations include diffraction limits for light microscopy, preparation artifacts, and potential sample damage. Developments continue in super-resolution techniques (STED, PALM/STORM), correlative and automated imaging, and digital analysis to increase speed and quantitative insight.
