IGFRezeptoren

IGFRezeptoren, or Insulin-like Growth Factor Receptors, are a family of transmembrane receptor tyrosine kinases. They are crucial for mediating the effects of insulin-like growth factors (IGFs), primarily IGF-1 and IGF-2. There are two main types of IGF receptors: the IGF-1 receptor (IGF-1R) and the IGF-2 receptor (IGF-2R).

The IGF-1 receptor is a well-characterized receptor that plays a vital role in cell growth, differentiation, proliferation, and survival. It is structurally similar to the insulin receptor. Upon binding of IGF-1 or IGF-2, the IGF-1R undergoes autophosphorylation, initiating a signaling cascade that involves the PI3K/Akt and MAPK pathways. These pathways regulate numerous cellular processes essential for development and tissue homeostasis. Dysregulation of IGF-1R signaling is implicated in various diseases, including cancer and diabetes.

The IGF-2 receptor, also known as the cation-independent mannose-6-phosphate receptor (CI-MPR), has a different function. While it can bind IGF-2, it does not possess intrinsic tyrosine kinase activity. Instead, its primary role is to act as a multifunctional receptor involved in lysosomal enzyme trafficking and endocytosis. It plays a role in clearing IGF-2 from circulation, thereby indirectly influencing IGF-1R signaling. The IGF-2R is essential for proper lysosomal enzyme targeting and the regulation of IGF-2 levels.
