GPC5

Glypican-5 (GPC5) is a member of the glypican family of heparan sulfate proteoglycans, proteins that influence cell signaling at the cell surface. Like other glypicans, GPC5 is tethered to the outer leaflet of the plasma membrane by a glycosylphosphatidylinositol (GPI) anchor and carries covalently attached heparan sulfate chains on its extracellular domain.

GPC5 is believed to function as a co-receptor or modulator of several signaling pathways, including Wnt, Hedgehog, and fibroblast growth factor (FGF) pathways. By binding ligands, presenting them to signaling receptors, or shaping the local extracellular matrix environment, GPC5 can influence processes such as cell proliferation, differentiation, and migration. Its activity is context-dependent and varies with tissue type and developmental stage.

GPC5 is expressed in multiple tissues and is regulated during development. In model systems, expression patterns have been observed in developing neural and mesenchymal tissues, among others, consistent with roles in patterning and growth. In adults, expression is more limited and can be altered in disease states.

Altered expression or function of GPC5 has been reported in various cancers and other diseases, and the protein is under investigation as a potential biomarker or therapeutic target. Research continues to clarify its precise roles in signaling networks and how its dysregulation contributes to pathology.
