FluorochromSig is a framework and database for cataloging the fluorescence signatures of fluorophores used in biological imaging. It seeks to standardize the collection and representation of spectral properties to support reliable multiplexing, spectral unmixing, and instrument calibration across platforms.

At its heart, FluorochromSig treats a fluorophore's signature as a vector of data that can vary with excitation wavelength, emission bandwidth, detector sensitivity, and environmental context. The data model records excitation and emission spectra, peak positions, bandwidth, relative brightness, quantum yield, photostability, and notes on environmental sensitivity (pH, ionic strength, temperature). Each signature is linked to metadata such as instrument settings, filter sets, and calibration references.

History and scope: The concept emerged from needs in flow cytometry, confocal and wide-field imaging to compare data across instruments. A community consortium established a shared schema and an open database to host fluorophore signatures, with ongoing updates as new dyes and measurement conditions arise.

Applications: Researchers use FluorochromSig to design high-plex panels, perform spectral unmixing in hyperspectral imaging, compare fluorophores from different vendors, and validate instrument performance. The framework also aids reproducibility by providing standardized reference signatures.

Limitations and future directions: Signatures can shift with sample context, staining protocols, and illumination power, so signatures are best used with explicit metadata.
