Exonucleolytische refers to a process involving enzymes called exonucleases. These enzymes degrade nucleic acid molecules, such as DNA or RNA, by sequentially removing nucleotides from one of the ends. This is in contrast to endonucleases, which cleave nucleic acids internally.

There are two main types of exonucleases: 5' exonucleases and 3' exonucleases. A 5' exonuclease will remove nucleotides starting from the 5' end of the nucleic acid strand, while a 3' exonuclease will remove them from the 3' end. Some exonucleases are specific to DNA, while others act on RNA.

For instance, some antiviral drugs work by inhibiting viral exonucleases, thereby preventing the replication of viral genetic material.
