DnaGprimasi

DnaGprimase is an essential enzyme in DNA replication in bacteria. It is a type II DNA topoisomerase that synthesizes short RNA primers. These RNA primers are crucial because DNA polymerases, the enzymes responsible for synthesizing new DNA strands, cannot initiate DNA synthesis de novo; they require a pre-existing 3'-hydroxyl group to add nucleotides to. DnaGprimase provides this necessary starting point by creating these short RNA sequences.

The primase enzyme is a type of RNA polymerase, meaning it uses a DNA template to synthesize RNA. Specifically, DnaG primase recognizes and binds to specific DNA sequences known as primase recognition sites, which are often located near the origin of replication. Once bound, it synthesizes an RNA oligomer, typically 10-60 nucleotides in length, which is complementary to the DNA template strand. This RNA primer is then extended by DNA polymerase III, which adds deoxyribonucleotides to the 3' end of the primer, creating a new DNA strand.

In bacteria, DnaG primase plays a critical role in both leading and lagging strand synthesis. On the leading strand, a single primer is synthesized at the origin of replication to initiate continuous DNA synthesis. On the lagging strand, which is synthesized discontinuously, DnaG primase must repeatedly synthesize short RNA primers at intervals along the template as the replication fork progresses. These primers are later removed and replaced with DNA by other enzymes, and the resulting Okazaki fragments are ligated together. The activity of DnaG primase is tightly regulated to ensure accurate and timely initiation of DNA replication.
