DNA is constantly exposed to various damaging agents, including environmental factors like radiation and chemicals, as well as errors that occur during normal cellular processes like DNA replication. If left unrepaired, these DNA lesions can lead to mutations, genomic instability, and ultimately cell death or disease, such as cancer.

The DNA damage response system is a multi-faceted network involving a multitude of proteins and pathways. Upon sensing DNA damage, cells trigger signaling cascades that halt the cell cycle to prevent the replication of damaged DNA. This pause allows time for repair mechanisms to act. There are several distinct DNA repair pathways, each specialized to address different types of DNA lesions.
