DNA methyltransferase, often abbreviated as DNMT, refers to a group of enzymes that catalyze the transfer of a methyl group to a DNA molecule. This process, known as DNA methylation, is a fundamental epigenetic modification that plays a crucial role in regulating gene expression. DNMTs are essential for various cellular processes, including development, differentiation, and maintaining genomic stability.

The primary function of DNA methyltransferases is to add a methyl group to the cytosine base, typically within a CpG dinucleotide context, although other methylation patterns exist. This methylation event can alter the physical structure of DNA and influence the binding of proteins, thereby affecting whether a gene is transcribed or silenced. There are several known DNMT enzymes in mammals, including DNMT1, DNMT3A, and DNMT3B, each with distinct roles in establishing and maintaining methylation patterns.

DNMT1 is primarily responsible for maintaining existing DNA methylation patterns during DNA replication, ensuring that methylation is passed on to daughter cells. In contrast, DNMT3A and DNMT3B are de novo methyltransferases, meaning they establish new methylation patterns during development or in response to environmental cues. Aberrant DNA methylation, often involving either hypermethylation or hypomethylation of specific genes, is implicated in a wide range of diseases, including cancer, neurological disorders, and autoimmune conditions. Understanding the activity and regulation of DNA methyltransferases is therefore of significant interest in both basic research and the development of therapeutic strategies.
