Crossmatching is a laboratory procedure used to test the compatibility of blood or tissues between two individuals. This process is crucial for ensuring safe transplantation and transfusion. The main goal of crossmatching is to identify whether the immune system of one individual will react adversely to the cells or tissues of another.

The crossmatching process typically involves taking a blood sample from the recipient and an individual whose cells or tissue will be transplanted or transfused. The samples are then sent to a laboratory where the immune system's reaction against the donor's cells is tested. This is done using various tests such as the major histocompatibility complex (MHC) antigens, cytotoxicity assays, and flow cytometry.

The tests evaluate the recipient's immune system's ability to recognize and attack the donor's cells as foreign. The goal is to identify if the recipient's immune system has pre-existing antibodies against the donor's cells, which can lead to a severe reaction during transplantation or transfusion.

False-negative crossmatch results can occur if the recipient's immune system has a low level of pre-existing antibodies against the donor's cells.
