Aptazymes

Aptazymes, also known as catalytic aptamers, are single-stranded nucleic acid molecules, typically RNA or DNA, that possess enzymatic activity. Unlike protein enzymes, aptazymes are created through in vitro selection processes, known as SELEX (Systematic Evolution of Ligands by Exponential Enrichment), where a combinatorial library of nucleic acid sequences is screened for the desired catalytic function. These sequences fold into specific three-dimensional structures that enable them to bind to a target molecule and catalyze a specific chemical reaction.

The catalytic activity of aptazymes can range widely, including the cleavage of other nucleic acid molecules (ribozymes and deoxyribozymes), ligation reactions, phosphorylation, and even the synthesis of peptide bonds. The specificity and catalytic efficiency of aptazymes can be fine-tuned through further rounds of selection and modification. Their ability to perform catalysis, combined with the relative ease of their in vitro generation and their stability, makes them attractive for a variety of applications.
