Allyyliamiinilääke refers to a class of medications that contain an allylamine functional group. This structural feature is common in several antifungal drugs, particularly those used topically and orally. The primary mechanism of action for allylamine antifungals involves the inhibition of squalene epoxidase, a key enzyme in the fungal ergosterol biosynthesis pathway. Ergosterol is an essential component of fungal cell membranes, analogous to cholesterol in mammalian cells. By blocking the synthesis of ergosterol, allylamine drugs disrupt the integrity and function of the fungal cell membrane, leading to cell death.

The most well-known allylamine antifungal is terbinafine, available in both topical and oral formulations. Other examples include naftifine and amorolfine, which are primarily used topically. These medications are effective against a broad spectrum of fungi, including dermatophytes, which are responsible for common fungal infections of the skin, hair, and nails such as athlete's foot, ringworm, and onychomycosis. While generally well-tolerated, allylamine antifungals can have side effects, which may vary depending on the specific drug and route of administration. Oral terbinafine, for instance, can rarely cause liver problems, and topical formulations may cause local irritation. Research continues to explore the efficacy and potential of allylamine derivatives in combating emerging fungal resistance.
