Ribulose-5-phosphate is an intermediate compound in the metabolic pathways of many organisms, playing a central role in both the Calvin cycle (a key process in photosynthesis) and the pentose phosphate pathway (PPP). It is a phosphorylated derivative of ribulose, a five-carbon sugar, and serves as a precursor for the synthesis of various essential biomolecules.

In the Calvin cycle, ribulose-5-phosphate is generated during the regeneration phase, where it is converted from ribulose-1,5-bisphosphate (RuBP) after carbon fixation. RuBP reacts with carbon dioxide in a reaction catalyzed by the enzyme RuBisCO, forming an unstable six-carbon intermediate that splits into two molecules of 3-phosphoglycerate. Through a series of enzymatic steps, these molecules are eventually converted into ribulose-5-phosphate, enabling the cycle to continue.

The pentose phosphate pathway also involves ribulose-5-phosphate, though its role differs slightly. In this pathway, glucose-6-phosphate is oxidized and decarboxylated to produce ribulose-5-phosphate, which can then be rearranged into other pentose sugars like ribose-5-phosphate, a critical precursor for nucleotide synthesis. Ribulose-5-phosphate can also be converted into glycolytic intermediates, such as glyceraldehyde-3-phosphate, to supply energy and biosynthetic precursors.
