Protongradientti refers to a difference in proton concentration across a biological membrane. This difference creates an electrochemical gradient, which is a form of potential energy. The gradient is established by proton pumps, which actively transport protons from one side of the membrane to the other, often utilizing energy derived from electron transport chains or ATP hydrolysis.

The most well-known example of a proton gradient is found in mitochondria during cellular respiration and in chloroplasts during photosynthesis. In these organelles, the proton gradient across the inner mitochondrial membrane or the thylakoid membrane, respectively, drives ATP synthesis. As protons flow back across the membrane down their electrochemical gradient through a specialized enzyme called ATP synthase, the energy released is used to produce ATP, the cell's primary energy currency. This process is known as chemiosmosis. Proton gradients are also crucial for other cellular functions, including nutrient transport and maintaining cell volume. The dissipation of this gradient represents a loss of potential energy that can be harnessed for work.
