For proteins requiring multiple subunits, these folded chains then associate with each other, driven by various non-covalent interactions such as hydrogen bonds, ionic bonds, and hydrophobic interactions, as well as sometimes covalent disulfide bonds.

The quaternary structure of a protein describes the spatial arrangement of its subunits. This arrangement dictates the protein's overall shape and the accessibility of its active sites or interaction surfaces. For instance, hemoglobin, a protein responsible for oxygen transport, is a heterotetramer, consisting of two alpha-globin subunits and two beta-globin subunits. The precise assembly of these subunits allows hemoglobin to efficiently bind and release oxygen. Variations in subunit composition can lead to proteins with different functions or properties, even if they share a common ancestral origin.
