Inositoltrisphosphate, often abbreviated as IP3, is a signaling molecule within cells. It is derived from the membrane phospholipid phosphatidylinositol 4,5-bisphosphate (PIP2). The formation of IP3 is a crucial step in a common cellular signaling pathway known as the inositol trisphosphate pathway.

This pathway is activated by various extracellular signals, such as hormones and neurotransmitters, that bind to specific receptors on the cell surface. This binding event triggers the activation of an enzyme called phospholipase C. Phospholipase C then cleaves PIP2 into two second messengers: IP3 and diacylglycerol (DAG).

The primary role of IP3 is to mobilize intracellular calcium ions. Upon its formation, IP3 diffuses through the cytoplasm and binds to specific receptors located on the endoplasmic reticulum, a cellular organelle that stores calcium. This binding causes the opening of calcium channels on the endoplasmic reticulum, leading to a rapid release of stored calcium into the cytosol.

The increase in cytosolic calcium concentration then acts as a signal to activate a multitude of downstream cellular processes. These can include muscle contraction, neurotransmitter release, gene expression, and cell growth, depending on the specific cell type and the initiating signal. IP3 is a transient molecule, and its effects are typically terminated by its dephosphorylation, converting it back into less active forms.
