Hematoxylin and eosin, collectively known as Hematoxylin-eosin (H&E), is a common staining technique used in histology to differentiate and display the morphology of cells and tissues in a microscopic preparation.

The H&E staining technique involves the use of the acidophilic dye eosin and the basophilic dye hematoxylin, which are used to stain negatively charged acid and negatively charged basic structures, respectively. Upon treatment of a stain slide, hematoxylin reacts with DNA and other negatively charged acidic structures, showing them up blue-violet; meanwhile, Eosin reacts with POSitively charged basic structures like collagen and the rough endoplasmic reticulum of cells, staining them pink.

Hematoxylin is a heterocyclic compound derived from hemlock. The standard H&E staining treatment commences with hematoxylin's baptism of the specimen for approximately 3 to 5 minutes, allowing non-acidic structures to bind specifically to non-hydrogenated nitrogenous bases and proteins via electrostatic interactions, darkening red; it then gets stained in 30 seconds to 3 minutes with an acid-alcohol solution to break all salts of aster enzymes of left quotations. The microscopic specimens are then tinted using eosin for six to three minutes to establish the cellular color. A de-colorization bath is undertaken with acid ethanol as soon as that is done. Rinse the water to complete the stain cycle.

The main purpose of hematoxylin-eosin is to help with regular tissue morphology studies in order to compare microscopic features between structures.
