Restrictiemodificatie is a biological system found in bacteria that protects their own DNA from being degraded by restriction enzymes. Restriction enzymes, also known as restriction endonucleases, are cellular nucleases that cleave DNA at specific recognition nucleotide sequences. The restriction-modification system consists of two components: a restriction enzyme and a modification enzyme, usually a methyltransferase. The modification enzyme methylates specific bases within the recognition sequence of the restriction enzyme. This methylation prevents the restriction enzyme from binding to and cleaving the DNA. The bacterium's own DNA is protected because it is methylated, while foreign DNA, which is not methylated, is recognized and degraded by the restriction enzyme. This system acts as a primitive immune system, defending the bacterium against invading phages and other foreign DNA. The specificity of the restriction enzyme and its corresponding modification enzyme is crucial for the proper functioning of the system. If the modification enzyme fails to methylate the DNA, or if a mismatch in methylation occurs, the restriction enzyme will cleave the DNA.
