RNAprimers are short sequences of RNA that are essential for DNA replication. They serve as a starting point for DNA polymerase, the enzyme responsible for synthesizing new DNA strands. DNA polymerase cannot initiate DNA synthesis on its own; it requires a pre-existing nucleotide to add to. RNAprimers provide this crucial starting point.

In the context of DNA replication, an enzyme called primase synthesizes these short RNA sequences, typically around 5-10 nucleotides long, complementary to the DNA template strand. Once the RNA primer is synthesized, DNA polymerase can bind to it and begin adding DNA nucleotides, extending the new DNA strand in the 5' to 3' direction.

On the leading strand, which is synthesized continuously, only one RNA primer is needed at the very beginning of replication. However, on the lagging strand, which is synthesized discontinuously in fragments called Okazaki fragments, multiple RNA primers are required. Each Okazaki fragment is initiated by an RNA primer.

After the DNA polymerase has synthesized the DNA portion of the Okazaki fragment, the RNA primer must be removed and replaced with DNA. This is typically done by another DNA polymerase or an enzyme with exonuclease activity, which removes the RNA nucleotides. Finally, DNA ligase seals the gaps between the DNA fragments, creating a continuous DNA strand. RNAprimers are thus indispensable for the accurate and complete duplication of the genome.
