Therefore, PDE3 inhibition can result in widening of blood vessels. In platelets, PDE3 contributes to the regulation of platelet aggregation. Inhibiting PDE3 can reduce platelet clumping, which has implications for preventing blood clots.

Several drugs target PDE3 enzymes, acting as inhibitors. These inhibitors are used clinically for conditions such as heart failure, where they can improve cardiac output, and in some cases, for peripheral artery disease.
