Notch signalveje refer to a fundamental cell-cell communication system found in multicellular organisms. This pathway plays a crucial role in a wide range of developmental processes, including cell fate determination, tissue patterning, and stem cell maintenance. The Notch pathway is activated when a transmembrane receptor, known as the Notch receptor, binds to a transmembrane ligand on an adjacent cell. This binding event triggers a series of proteolytic cleavages of the Notch receptor.

The most significant cleavage event releases the Notch intracellular domain (NICD). This NICD then translocates into the nucleus, where it interacts with a transcription factor called Suppressor of Hairless (Su(H)) in Drosophila, or CSL (CBF1/Su(H)/Lag-1) in mammals. This interaction converts Su(H)/CSL from a transcriptional repressor into a transcriptional activator. The activated complex then recruits co-activators, leading to the transcription of specific target genes. These target genes encode proteins that influence cell proliferation, differentiation, and apoptosis.

The Notch pathway is highly conserved across species, highlighting its importance in biological systems. Dysregulation of Notch signaling has been implicated in various diseases, including cancer, developmental disorders, and autoimmune conditions. Due to its critical roles, the Notch pathway is an active area of research, with ongoing efforts to understand its complex mechanisms and explore its therapeutic potential.
